A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Binding of tesamorelin to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and the release of growth hormone into circulation. Because the peptide acts upstream of the growth hormone axis, its effects are partly mediated by hepatic insulin-like growth factor 1 (IGF-1) production. The pulsatile character of endogenous growth hormone secretion is preserved rather than replaced. Whether amplified signaling produces effects beyond those of native GHRH remains an area of ongoing investigation.

Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
== History == The predecessor of modern countercurrent chromatography theory and practice was countercurrent distribution (CCD). The theory of CCD was described in the 1930s by Randall and Longtin. Archer Martin and Richard Laurence Millington Synge developed the methodology further during the 1940s. Finally, Lyman C. Craig introduced the Craig countercurrent distribution apparatus in 1944 which made CCD practical for laboratory work. CCD was used to separate a wide variety of useful compounds for several decades.
Other nanoparticles are polymer-based, meaning they are made from a natural polymer such as polylactic acid (PLA), polyglycolide (PLG), polylactide-co-glycolide (PLGA), and polycyanoacrylate (PCA). Some studies have found that polymeric nanoparticles may provide better results for drug delivery relative to lipid-based nanoparticles because they may increase the stability of the drugs or proteins being transported. Polymeric nanoparticles may also contain beneficial controlled release mechanisms.
== History == The SIRIUS software is developed by the group of Sebastian Böcker at the Friedrich Schiller University Jena, Germany and since 2019 together with Bright Giant GmbH. SIRIUS development started in 2009 as a software for identification of the molecular formula by decomposing high-resolution isotope patterns (also called MS1 data). The name is an akronym resulting from this original purpose: Sum formula Identification by Ranking Isotope patterns Using mass Spectrometry. In 2008 the group introduced the concept of fragmentation trees for identification of the molecular formula based on fragmentation mass spectrometry data, also called tandem MS or MS2 data. Back then, identification of small molecules was approached by searching in a reference spectral library. Examples of such libraries include MassBank, METLIN, or NIST/EPA/NIH EI-MS Library. However, this is limited to known molecules with available standards that have been measured and put in a reference spectral library. For unknown molecules, identification of the molecular formula is a crucial step. In 2011/2012, the group conceived fragmentation trees as a means of structural elucidation by automatically comparing these fragmentation trees. Fragmentation pattern similarities are strongly correlated with the chemical similarity of molecules. Thus, aligning the fragmentation tree of an unknown molecule to a set of known molecules helps to elucidate its structure. Fragmentation trees were introduced in SIRIUS 2.
=== Legal status === In January 2016, Eli Lilly submitted a new drug application to the US Food and Drug Administration (FDA) for the approval of baricitinib to treat moderately-to-severely active rheumatoid arthritis. In December 2016, the Committee for Medicinal Products for Human Use (CHMP) of the European Medicines Agency (EMA) recommended the approval of baricitinib as a therapy for rheumatoid arthritis. European Union approval was granted in February 2017. Despite widespread expectations that the FDA would approve baricitinib for rheumatoid arthritis, in April 2017, the FDA issued a rejection, citing concerns about dosing and safety. In May 2018, baricitinib was approved in the United States for the treatment of rheumatoid arthritis. In March 2020, the US FDA granted breakthrough therapy designation to baricitinib for the treatment of alopecia areata and granted approval in June 2022. The efficacy and safety of baricitinib in alopecia areata was studied in two randomized, double-blind, placebo-controlled trials (Trial AA-1 and Trial AA-2) with participants who had at least 50% scalp hair loss as measured by the Severity of Alopecia Tool (SALT) for more than six months. Participants in these trials received either a placebo, 2 milligrams of baricitinib, or 4 milligrams of baricitinib every day. The primary measurement of efficacy for both trials was the proportion of participants who achieved at least 80% scalp hair coverage at week 36.
Bean chips are chips prepared using beans as a primary ingredient. Bean chips have been described as being higher in fiber and protein compared to corn and potato chips. Bean chips may be prepared from a variety of bean types and rice, such as brown rice, pinto beans, black beans, and white beans. Some bean chips are prepared using green beans. They may be prepared from a bean dough that is steamed, sliced, and fried. They may be consumed as a snack food and may be accompanied by various dips.

=== Islamism === Hitchens was deeply shocked by 14 February 1989 fatwa against his longtime friend Salman Rushdie. He became increasingly critical of what he called "theocratic fascism" or "fascism with an Islamic face": radical Islamists who supported the fatwa against Rushdie and sought the recreation of the medieval caliphate. In 2007, Hitchens told New York magazine "If you ask specifically what is wrong with Islam, it makes the same mistakes as the preceding religions, but it makes another mistake, which is that it’s unalterable. You notice how liberals keep saying, “If only Islam would have a Reformation"—it can't have one. It says it can't. It's extremely dangerous in that way". Hitchens is often credited with coining the term "Islamofascism", but Hitchens himself denied it, attributing its coinage to Malise Ruthven. In February 2006, Hitchens helped organize a pro-Denmark rally outside the Danish Embassy in Washington, DC, in response to the Jyllands-Posten Muhammad cartoons controversy. Hitchens has received criticism for his attacks on Islam and has been referred to as Islamophobic. Hitchens has called Islamophobia "A stupid term—Islamophobia—has been put into circulation to try and suggest that a foul prejudice lurks behind any misgivings about Islam's infallible "message".

=== United States === The Centers for Disease Control and Prevention (CDC) issued a health alert to report that between March 2013 and May 2013, 14 overdose deaths related to injected acetylfentanyl had occurred among intravenous drug users (ages between 19 and 57 years) in Rhode Island. After confirming five overdoses in one county, including a fatality, Pennsylvania asked coroners and medical examiners across the state to screen for acetylfentanyl. As a result of this investigation, Pennsylvania confirmed at least one acetylfentanyl overdose death and attributed at least 50 fatalities to either fentanyl or acetylfentanyl during the first half of 2013. In July 2015, the DEA informed about 52 confirmed fatalities involving acetylfentanyl in the United States between 2013 and 2015.
Bicalutamide monotherapy has been found to decrease circulating levels of prostate-specific antigen (PSA), a marker of prostate cancer growth, by 57% at 10 mg/day, 73% at 30 mg/day, 90% at 50 mg/day, 97% at 100 mg/day, and 97% at 150 mg/day, while a 97% reduction in PSA is observed with 50 mg/day bicalutamide as a part of CAB. It has also been reported that bicalutamide monotherapy decreases median circulating levels of PSA at 3 months by 86.7% at 100 mg/day, 91.1% at 150 mg/day, and 93.8% at 200 mg/day (relative to 94–97% for castration). Above a bicalutamide monotherapy dosage of 200 mg/day, up to 600 mg/day, decreases in PSA levels reach a plateau. In a study of very-high-dose bicalutamide monotherapy, decreases in PSA levels after 12 weeks were approximately 93% with 300 mg/day, 96% with 450 mg/day, 96% with 600 mg/day, and 96% with castration. Earlier studies with bicalutamide instead assessed changes in prostatic acid phosphatase (PAP) levels; proportions of patients with decreases of PAP of greater than or equal to 50% were 33% with 10 mg/day, 53% with 30 mg/day, and 83% with 50 mg/day bicalutamide. PSA is a more sensitive and specific prostate cancer tumor marker than PAP and subsequent studies employed PSA. Despite the high medication levels that are achieved, due to their relatively low affinities for the AR, it has been suggested that 5 to 10% of DHT may remain unblocked in the prostate gland with CAB using standard doses of first-generation NSAAs.
== History == Fenfluramine as a single drug was first introduced in the 1970s, but was not popular because it only temporarily reduced weight. A 1984 study found a weight loss of 7.5 kg on average in 24 weeks, as compared to 4.4 kg under placebo. It sold modestly until the 1990s, when it was combined with phentermine and heavily marketed.

It mirrors the 44-residue form of human growth hormone-releasing hormone. A hexenoyl group on the N-terminal tyrosine distinguishes it from the unmodified hormone. The change is intended to improve resistance to enzymatic breakdown.
