=== Approximation of the reactant === During enzyme catalytic reaction, the substrate and active site are brought together in a close proximity. This approach has various purposes. Firstly, when substrates bind within the active site the effective concentration of it significantly increases than in solution. This means the number of substrate molecules involved in the reaction is also increased. This process also reduces the desolvation energy required for the reaction to occur. In solution substrate molecules are surrounded by solvent molecules and energy is required for enzyme molecules to replace them and contact with the substrate. Since bulk molecules can be excluded from the active site this energy output can be minimised. Next, the active site is designed to reorient the substrate to reduce the activation energy for the reaction to occur. The alignment of the substrate, after binding, is locked in a high energy state and can proceed to the next step. In addition, this binding is favoured by entropy as the energy cost associated with solution reaction is largely eliminated since solvent cannot enter active site. In the end, the active site may manipulate the Molecular orbital of the substrate into a suitable orientation to reduce activation energy. The electrostatic states of substrate and active site must be complementary to each other. A polarized negatively charged amino acid side chain will repel uncharged substrate. But if the transition state involves the formation of an ion centre then the side chain will now produce a favourable interaction.

=== Cancer === Based on cell growth experiments, animal cancer models, and epidemiological studies, it appears that IGFBP-3 functions as a low-penetrance tumor suppressor gene. Dysregulation of IGFBP-3 has been implicated in many cancers. Downregulation of its tissue expression by promoter hypermethylation in some cancers, such as hepatoma and non-small cell lung cancer may be associated with poor patient outcome. However, consistent with the dual inhibitory and stimulatory roles of IGFBP-3 seen in cell culture, there are other cancer types, such as breast cancer, pancreatic cancer, and clear cell renal cell cancer in which high tissue IGFBP-3 expression has been linked to poor prognostic features or patient outcome. The mechanisms regulating these contrasting effects of IGFBP-3 in vivo are not well understood. Since IGFBP-3 is abundant in the bloodstream of healthy adults (typically 2–4 mg/L), and is largely stabilized by its complex formation with IGFs and ALS, it is unlikely that tumor-derived IGFBP-3 has a large influence on circulating levels. There have been many studies linking circulating IGFBP-3 levels to the presence, or risk, of various cancers, or to patient outcomes. but unequivocal conclusions have often been lacking. For example, high plasma IGFBP-3 levels were associated with a reduced prospective risk of colorectal cancer in women. but in a study including men and women, colon cancer risk was positively associated with plasma IGFBP-3, while there was no significant association for rectal cancer.

Tucci and Platt developed the characters while working on a play at Yale University in 1988, with Tucci later completing the screenplay and directing the film. In 1999, Platt played the wealthy and eccentric crocodile enthusiast Hector in David E. Kelley's Lake Placid, alongside Bill Pullman and Bridget Fonda. Platt described Hector as "pretty abrasive and obnoxious at times, but, I hope, he has a way of growing on you. === Biogeography and environmental gradients === Ectomycorrhizal fungi are found throughout boreal, temperate and tropical ecosystems, primarily among the dominant woody-plant-producing families. Many of the fungal families common in temperate forests (e.g. Russulaceae, Boletaceae, Thelephoraceae) are also widespread in the Southern Hemisphere and tropical dipterocarp forests: although the plant families are quite different in temperate and tropical forests, the ectomycorrhizal fungi are fairly similar. The types of EcM fungi are affected by soil types both in the field and in the lab. For most types of plants and animals, species diversity increases towards the equator. This is called the latitudinal gradient of diversity (LGD). In contrast, there is evidence that EcM fungi may be at maximum diversity in the temperate zone. If this is the case, it might be explained by one or more of the following hypotheses: 1) EcM fungi may have evolved at higher latitudes with Pinaceae hosts, and be less able to compete in tropical climates; 2) the plants EcMs use as hosts might be more diverse in temperate conditions, and the structure of the soil in temperate regions may allow for higher niche differentiation and species accumulation; and 3) tropical EcM hosts are spread out more sparsely in small isolated forest islands that may reduce the population sizes and diversity of EcM fungi.
