Leuprorelin, also known as leuprolide, is a manufactured version of a hormone used to treat prostate cancer, breast cancer, endometriosis, uterine fibroids, for early puberty, as part of transgender hormone therapy, or to perform chemical castration of violent sex offenders. It is given by injection into a muscle or under the skin. Leuprorelin is in the gonadotropin-releasing hormone (GnRH) analogue family of medications. It works by decreasing gonadotropins, thereby decreasing testosterone and estradiol. Common side effects include hot flashes, unstable mood, trouble sleeping, headaches, and pain at the site of injection. Other side effects may include high blood sugar, allergic reactions, and problems with the pituitary gland. Use during pregnancy may harm foetal development. Leuprorelin was patented in 1973 and approved for medical use in the United States in 1985. It is on the World Health Organization's List of Essential Medicines. It is sold under the brand name Lupron, among others.
== Signs and symptoms == The hallmark sign of muscle atrophy is loss of lean muscle mass. This change may be difficult to detect due to obesity, changes in fat mass or edema. Changes in weight, limb or waist circumference are not reliable indicators of muscle mass changes. The predominant symptom is increased weakness which may result in difficulty or inability in performing physical tasks depending on what muscles are affected. Atrophy of the core or leg muscles may cause difficulty standing from a seated position, walking or climbing stairs and can cause increased falls. Atrophy of the throat muscles may cause difficulty swallowing and diaphragm atrophy can cause difficulty breathing. Muscle atrophy can be asymptomatic and may go undetected until a significant amount of muscle is lost.

== Etiopathogenesis == Recent studies suggest that the pathogenesis of DM is driven by the pathogenic internalization of autoantibodies. Although these antibodies target intracellular proteins, evidence indicates that they can enter different cell types and disrupt the function of their target autoantigens causing inflammation and damage. For example, anti-Mi-2 autoantibodies bind PHD-containing proteins, including component of the NuRD complex, inducing derepression of multiple genes, and in anti-MDA5 dermatomyositis, autoantibodies activate MDA5 directly inducing the activation of type I interferon pathways. The type I interferon pathway is especially prominent in DM and has become a major therapeutic target. Clinical responses to JAK inhibitors, anti-IFNβ therapy, and agents targeting the interferon receptor support the importance of this pathway in disease activity. Conversely, a negative trial of complement inhibition in immune-mediated necrotizing myopathy has challenged earlier models in which complement-mediated muscle injury was considered central to that subtype. The characteristic pathological feature is perifascicular muscle involvement, often accompanied by vasculopathy. Plasma cells are found in close proximity to affected areas with high type I interferon expression and have been observed to externalize immunoglobulin heavy- and light-chain RNA into surrounding muscle cells, suggesting that this may be a potential mechanism for immunoglobulin entry into affected cell types shared across different forms of DM.
