Practical handling centers on limiting moisture, oxygen, and temperature excursions. Lyophilized material is generally held at or below minus twenty degrees Celsius, protected from light and kept sealed until use. Once reconstituted, solutions are typically kept cold and used within a short window because hydrolysis and microbial growth both accelerate in liquid form. Repeated freeze-thaw cycles are avoided, since they promote aggregation. Vial contents should be inspected for particulates and clarity before analysis, and working aliquots are prepared to reduce the number of times the stock is opened.
Quantitation of the peptide relies mainly on reversed-phase high-performance liquid chromatography with ultraviolet detection, typically at 214 nanometers, where the peptide bond absorbs. Identity is confirmed by mass spectrometry, most often electrospray ionization coupled to liquid chromatography, and by peptide mapping after enzymatic digestion. Because related impurities differ only slightly in sequence or modification, method development emphasizes resolution rather than speed. Purity is usually reported as a percentage of the main peak area, with individual impurities listed separately when they exceed a defined reporting threshold.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.

The hexenoyl cap slows the enzyme step that trims the amino terminus of native GHRH, the same step that shortens its active lifetime in circulation. As a result, the modified peptide persists longer in plasma than the unmodified hormone in side-by-side comparison. Receptor activity stays broadly comparable, because the added group sits away from the residues that contact the binding site. This combination, preserved receptor activity with reduced degradation, explains why the analog was developed instead of the native sequence.
Several compounds share the GHRH framework, including sermorelin, the shorter 1-29 fragment, and other analogs built on the full 1-44 chain. Naming follows a common convention: a stem that identifies the peptide plus a suffix marking analog status. Reports may describe tesamorelin by its sequence fragment, as a GHRH(1-44) analog, or by its amino-terminal modification. Indexing the compound therefore requires searching all of these forms, since some older literature predates the current international nonproprietary name.
Tesamorelin is a synthetic peptide built from 44 amino acids and classified with the growth hormone–releasing hormone family. Its sequence corresponds to the human GHRH(1-44) backbone, carrying one structural change at the amino terminus. That change is a trans-3-hexenoyl group placed where the natural peptide would have an unmodified end. The modification is the feature that separates the compound from the endogenous hormone in name, in stability, and in how it is handled in the laboratory.
The peptide is supplied as a lyophilized powder in single-use vials and is normally kept refrigerated between two and eight degrees Celsius, protected from light. Once dissolved, the solution is handled carefully because peptide bonds and the acyl modification can degrade under warm or alkaline conditions. Vials are inspected for cracks, and the powder is checked for color and uniformity before handling. Temperature excursions during shipping are a frequent reason for quality questions.
Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from truncated or oxidized forms. Mass spectrometry confirms the expected molecular weight, and peptide mapping after enzymatic digestion verifies the amino acid sequence. Water content is measured because residual moisture affects stability, and tests for aggregates or particulates are standard for injectable peptides. Circular dichroism can indicate whether the molecule has adopted an unexpected secondary structure in solution.
Research supply is often accompanied by a certificate of analysis listing chromatographic purity, mass confirmation, and storage conditions. Laboratories compare that document with an independent test when material is intended for bench work, since certificates describe a batch rather than an individual vial. Published studies usually state the source and purity of the peptide because small differences in purity can shift measured activity. Full analytical validation is rarely reported, which leaves batch-to-batch comparability an open question.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
On 30 August 2023, Health Secretary Steve Barclay announced that it would be upgraded to a statutory inquiry, enabling witnesses to be compelled to give evidence. The inquiry, chaired by Lady Justice Thirlwall, formally opened on 22 November 2023. The public hearings began on 10 September 2024. In a ruling on 29 May 2024, the chair determined that remote live viewing would be available to Core Participants, their legal representatives and the media, but not to the wider public. During the hearings, colleagues gave evidence describing Letby as "excited and gossipy" when discussing the death of an infant, saying she preferred caring for unwell babies, complained when moved to less acute areas, and expressed eagerness for the first death of an infant to "get it out of the way". Former Health Secretary Jeremy Hunt apologised to families during the 2025 hearings, saying the government had taken "too long" to act. Letby's lawyers and other supporters later asked for the inquiry to be paused while her application to the Criminal Cases Review Commission was considered. Thirlwall rejected the request in March 2025 and said she intended to deliver her report in November 2025. In April 2026, the inquiry said that work on the report was still ongoing and that a publication date could not be confirmed.

==== Television and film ==== Ice-T was prominently featured as both a rapper and a breakdancer in Breakin' 'n' Enterin' (1983), a documentary about the early West Coast hip-hop scene. Ice-T's first film appearances were in the motion pictures, Breakin' (1984), and its sequel, Breakin' 2: Electric Boogaloo (1984). These films were released before Ice-T released his first LP, although he appears on the soundtrack to Breakin'. He has since stated he considers the films and his own performance in them to be "wack". In 1991, he embarked on a serious acting career, portraying police detective Scotty Appleton in Mario Van Peebles' action thriller New Jack City, gang leader Odessa (alongside Denzel Washington and John Lithgow) in Ricochet (1991), gang leader King James in Trespass (1992), followed by a notable lead role performance in Surviving the Game (1994), in addition to many supporting roles, such as J-Bone in Johnny Mnemonic (1995), and the marsupial mutant T-Saint in Tank Girl (1995). He was also interviewed in the Brent Owens documentary Pimps Up, Ho's Down, in which he claims to have had an extensive pimping background before getting into rap. He is quoted as saying "once you max something out, it ain't no fun no more. I couldn't really get no farther." He goes on to explain his pimping experience gave him the ability to get into new businesses. "I can't act, I really can't act, I ain't no rapper, it's all game. I'm just working these niggas." Later he raps at the Players Ball.
Changes in the concentrations of ligands or substrates may cause a switch in a protein's function. For example, in the presence of high iron concentrations, aconitase functions as an enzyme while at low iron concentration, aconitase functions as an iron-responsive element-binding protein (IREBP) to increase iron uptake. Proteins may also perform separate functions through the use of alternative binding sites that perform different tasks. An example of this is ceruloplasmin, a protein that functions as an oxidase in copper metabolism and moonlights as a copper-independent glutathione peroxidase. Lastly, phosphorylation may sometimes cause a switch in the function of a moonlighting protein. For example, phosphorylation of phosphoglucose isomerase (PGI) at Ser-185 by protein kinase CK2 causes it to stop functioning as an enzyme, while retaining its function as an autocrine motility factor. Hence when a mutation takes place that inactivates a function of a moonlighting proteins, the other function(s) are not necessarily affected. The crystal structures of several moonlighting proteins, such as I-AniI homing endonuclease / maturase and the PutA proline dehydrogenase / transcription factor, have been determined. An analysis of these crystal structures has demonstrated that moonlighting proteins can either perform both functions at the same time, or through conformational changes, alternate between two states, each of which is able to perform a separate function.

Veins vary in size from the smallest post-capillary venules, and more muscular venules, to small veins, medium veins, and large veins. The thickness of the walls of the veins varies as to their location – in the legs the vein walls are much thicker than those in the arms. In the circulatory system, blood first enters the venous system from capillary beds where arterial blood changes to venous blood. Large arteries such as the thoracic aorta, subclavian, femoral and popliteal arteries lie close to a single vein that drains the same region. Other arteries are often accompanied by a pair of veins held in a connective tissue sheath. The accompanying veins are known as venae comitantes, or satellite veins, and they run on either side of the artery. When an associated nerve is also enclosed, the sheath is known as a neurovascular bundle. This close proximity of the artery to the veins helps in venous return due to the pulsations in the artery. It also allows for the promotion of heat transfer from the larger arteries to the veins in a counterflow exchange that helps to preserve normal body heat.

Experiment D-8 used five dosimeters to measure the radiation in the spacecraft environment. Of particular interest was the South Atlantic Anomaly. Experiment D-9 was an experiment in simple spacecraft navigation where the crew used a sextant to measure their position using the stars. The objective was to investigate the feasibility of using this technique for lunar flights on the Apollo program. Experiments S-5 and S-6 were both photography experiments where they used a 70-millimeter Hasselblad camera to photograph the weather and terrain below them. There were two medical experiments: M-3 and M-4. The first was a bungee cord that the crew used for exercise. They said, after the mission, that this got harder as the mission went on, though this may have been due to a lack of sleep. The second was the phonocardiogram experiment, which had sensors attached to their bodies that measured heartbeat rates, especially during liftoff, EVA, and reentry. There were four engineering experiments:
In August 2025, Newsom secured legislative approval of California Proposition 50, a state constitutional amendment to allow congressional redistricting. The goal was to switch seats from Republican to Democratic control in response to the 2025 Texas gerrymander that would add five new Republican seats. Voters approved the measure in November.
Work on the game began with a Kickstarter funding campaign in 2015, before the developers were acquired by Microsoft Studios in 2018, supporting the developers to work on a version for the Xbox One. We Happy Few received mixed reviews from critics.
Bukele has threatened incarcerated gang members. At the beginning of the crackdown, he tweeted that the government had seized incarcerated gang members' belongings, removed their mattresses, and rationed their food. Bukele also posted a video of prisoners sleeping on floors and complaining about a lack of food and sanitation. He threatened to deprive them of food entirely in April 2022 if the gangs attempted to retaliate against the crackdown, citing rumors about revenge killings. After members of Barrio 18 killed three police officers in Santa Ana in June 2022, Bukele said at a press conference that the gangs were "going to pay dearly" for the "ambush" against the police. The government began destroying gravestones belonging to deceased gang members in November 2022 to prevent them from becoming "shrines" and Bukele compared the gravestone destructions to denazification in post-World War II Germany. He warned Salvadoran parents to keep their children away from gangs, since they would lead to "prison or death". Shortly after the crackdown began, Bukele called for the construction of a new 20,000-inmate prison. He announced the construction in July 2022 of the 40,000-inmate Terrorism Confinement Center (CECOT) in Tecoluca, that would be one of the world's largest prisons. In February 2023, Bukele posted a video on Twitter of him and members of his cabinet touring the prison. It is staffed by 250 police officers and 600 soldiers, and covers 410 acres (170 ha).

Reversed-phase high-performance liquid chromatography with ultraviolet detection is the standard technique for purity and content. Mass spectrometry provides orthogonal confirmation of identity. The two are normally used together rather than in isolation.
Removing water slows hydrolysis and limits the mobility that drives aggregation. A dry powder is also less hospitable to microbial growth. These factors make cold storage of the solid form more forgiving than storage of a reconstituted solution.
Methionine oxidation, asparagine and glutamine deamidation, and non-covalent or covalent aggregation are the main routes reported for peptides of this class. Each is tracked as a separate impurity. Their relative abundance depends on formulation and storage history.
It is a synthetic peptide built from 44 amino acids arranged in the same order as human growth hormone-releasing hormone. A short fatty-acid chain, described as a trans-3-hexenoyl group, is attached to the first amino acid. The finished molecule is formulated as a sterile powder that is dissolved before use.
