Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Binding of tesamorelin to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and the release of growth hormone into circulation. Because the peptide acts upstream of the growth hormone axis, its effects are partly mediated by hepatic insulin-like growth factor 1 (IGF-1) production. The pulsatile character of endogenous growth hormone secretion is preserved rather than replaced. Whether amplified signaling produces effects beyond those of native GHRH remains an area of ongoing investigation.

Physicochemical behavior is dominated by the peptide backbone. The molecule is hydrophilic and carries a net positive charge near neutral pH, owing to several arginine and lysine residues. In solution it adopts a largely unstructured conformation, and aggregation is a known concern for peptide products of this size. Oxidation of methionine and deamidation of asparagine or glutamine residues are the principal chemical degradation routes. These liabilities shape how the material is formulated, handled, and analyzed, and they explain why lyophilized presentations are common in research settings.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, built from 44 amino acids. Its sequence follows the natural human GHRH(1-44) backbone, with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification blocks recognition by dipeptidyl peptidase IV, the enzyme that rapidly truncates the native hormone in circulation. The result is a molecule with a substantially longer plasma residence time than unmodified GHRH, which makes it practical for clinical and laboratory study.
Receptor-level activity begins when the peptide binds the GHRH receptor, a class B G-protein-coupled receptor found on pituitary somatotroph cells. Occupancy triggers Gs-mediated activation of adenylyl cyclase and a rise in intracellular cyclic AMP, which in turn promotes synthesis and pulsatile release of growth hormone. Because the compound acts upstream of the growth hormone axis rather than supplying hormone directly, its effect depends on intact pituitary function. Binding studies in cell culture and animal models have established this pathway; the detailed kinetics of receptor recycling in humans remain less well characterized.
Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
=== Tesla Cybertruck incident === On December 24, 2025, a video from a livestream of Peters hitting a man with a Tesla Cybertruck in Miami-Dade County quickly circulated online. In the video, the man climbs onto the hood of the car, and someone off-camera encourages Peters to start driving. He accelerates and appears to run over the man before asking if he is dead. A woman sitting next to him says, "I don't know," to which he responds, "Hopefully." Later in the livestream, Peters speaks to a man in a reflective vest and states that he hit the man in self-defense. Peters alleges that the man had been stalking the streamer and had previously thrown red liquid on him; he also alleges that there were multiple people, one of whom looked like they "had a pistol" underneath their clothing, surrounding his car. A call between him and fellow livestreamer Adin Ross, in which Ross advised him to keep livestreaming and refrain from speaking on the situation, also circulated online. Williams, writing for The Atlantic, called the livestream "perhaps [Peters's] most viral moment". Following the incident, he was banned from Kick. James Fishback, then a fringe candidate for the Republican nomination in the 2026 Florida gubernatorial election, tweeted that Peters had done "nothing wrong" by hitting the man with his car.

The preconditions to the development of a living cell like the LUCA are known, though disputed in detail: a habitable world is formed with a supply of minerals and liquid water and sources of free energy inflows such as our Sun and geothermal heat. Prebiotic synthesis creates a range of simple organic compounds, which are assembled into polymers such as proteins and RNA. On the other side, the process after the LUCA is readily understood: biological evolution caused the development of a wide range of species with varied forms and biochemical capabilities. However, the derivation of the LUCA from simple components is far from understood. Although Earth remains the only place where life is known, the science of astrobiology seeks evidence of life on other planets. The 2015 NASA strategy on the origin of life aimed to solve the puzzle by identifying interactions, intermediary structures and functions, energy sources, and environmental factors that contributed to evolvable macromolecular systems, and mapping the chemical landscape of potential primordial informational polymers. The advent of such polymers was most likely a critical step in prebiotic chemical evolution. Those polymers derived, in turn, from simple organic compounds such as nucleobases, amino acids, and sugars, likely formed by reactions in the environment. A successful theory of the origin of life must explain how all these chemicals came into being.

This final structure will form several distinct layers of the articular cartilage found in all synovial joints including the deep zone (closest to the bone), middle zone, and superficial zone (closest to the synovial fluid). Maintenance of articular cartilage is guided by a balance of anabolic (cartilage generating) and catabolic (cartilage degrading factors), in a manner similar to the maintenance of bone. Over the lifetime of the organism, anabolic factors and catabolic factors are generally in balance, however, as the organism ages, catabolism predominates and cartilage begins to degrade. Eventually, the loss of hyaline cartilage matrix and reduction in the chondrocyte content of the hyaline cartilage matrix results in the development of joint disease such as osteoarthritis. Overexpression of hyaline-cartilage specific anabolic factors, such as FGF18, appears to restore the balance between cartilage loss and generation.
The articular cartilage function is dependent on the molecular composition of the extracellular matrix (ECM). The ECM consists mainly of proteoglycan and collagens. The main proteoglycan in cartilage is aggrecan, which, as its name suggests, forms large aggregates with hyaluronan and with itself. These aggregates are negatively charged and hold water in the tissue. The collagen, mostly collagen type II, constrains the proteoglycans. The ECM responds to tensile and compressive forces that are experienced by the cartilage. Cartilage growth thus refers to the matrix deposition, but can also refer to both the growth and remodeling of the extracellular matrix. Due to the great stress on the patellofemoral joint during resisted knee extension, the articular cartilage of the patella is among the thickest in the human body. The ECM of articular cartilage is classified into three regions: the pericellular matrix, the territorial matrix, and the interterritorial matrix.
In some cases, even marine taxa that had acquired spongy bone can revert to thicker, compact bones if they become adapted to live in shallow water, or in hypersaline (denser) water. Many animals, particularly herbivores, practice osteophagy—the eating of bones. This is presumably carried out in order to replenish lacking phosphate. Many bone diseases that affect humans also affect other vertebrates—an example of one disorder is skeletal fluorosis.
=== Other === Cosmetics; Medical makeup can temporarily conceal scars. This is most commonly used for facial scars. Dermabrasion involves the removal of the surface of the skin with special equipment, and usually involves a local anaesthetic. A 2012 literature review found weak evidence that massage was efficacious in scar management. Any beneficial effect appeared to be greater in wounds created by surgical incision than for traumatic or burn wounds. A 2022 scoping review covering twenty-five studies of 1515 participants reported that all studies reviewed reported favorable outcomes for scar massage, but that "while there may be benefits to scar massage in reducing pain, increasing movement and improving scar characteristics", there was a lack of "consistent research methods, intervention protocols and outcome measures". The temporarily high level of 135Xe with its high neutron absorption cross-section makes it difficult to restart the reactor for several hours. The neutron-absorbing 135Xe acts like a control rod, reducing reactivity. The inability of a reactor to be started due to the effects of 135Xe is sometimes referred to as xenon-precluded start-up, and the reactor is said to be "poisoned out". The period of time that the reactor is unable to overcome the effects of 135Xe is called the "xenon dead time". If sufficient reactivity control authority is available, the reactor can be restarted, but the xenon burn-out transient must be carefully managed. As the control rods are extracted and criticality is reached, neutron flux increases many orders of magnitude and the 135Xe begins to absorb neutrons and be transmuted to 136Xe. The reactor burns off the nuclear poison. As this happens, the reactivity and neutron flux increases, and the control rods must be gradually reinserted to counter the loss of neutron absorption by the 135Xe. Otherwise, the reactor neutron flux will continue to increase, burning off even more xenon poison, on a path to runaway criticality. The time constant for this burn-off transient depends on the reactor design, power level history of the reactor for the past several days, and the new power setting. For a typical step up from 50% power to 100% power, 135Xe concentration falls for about 3 hours.

Senator Andy Kim of New Jersey described it as "unhinged and embarrassing", and Chris Murphy, a member of the Senate Foreign Affairs Committee, as "the ramblings of a man who has lost touch with reality". Vin Gupta, the medical analyst for NBC News, said the letter "crossed a line of proper adult behavior" and should have had a "more thorough public assessment of his neurological fitness," stating that Trump's behavior including his letter to Støre could be signs of early Alzheimer's or frontotemporal dementia. On January 21, 2026, while speaking in Davos, Trump would at least four times refer to Greenland as "Iceland." On January 22, 2026, Trump spoke before world leaders at a summit in Davos, and photographers captured photos of him with a large bruise on his left hand, unlike the bruise on his other hand, which the administration had to address publicly. Trump has publicly attributed the bruising to shaking hands, but he is accustomed to using his right hand, not his left. On January 29, 2026, shortly after the signing of Executive Order 14379[ws] "Addressing Addiction through the Great American Recovery Initiative", Trump and his cabinet dismissed news reporters from the Oval Office without an interview. Some social media users, including Rebekah Jones, speculated that the press conference ended abruptly due to Trump defecating himself. These accusations cited an unidentified sound and apparent reactions from Pam Bondi and Doug Burgum's wife Kathryn heard and seen in Forbes's live coverage.
The trial, carried out between 1946 and 1947, aimed to test the efficacy of the chemical streptomycin for curing pulmonary tuberculosis. The trial was both double-blind and placebo-controlled. The methodology of clinical trials was further developed by Sir Austin Bradford Hill, who had been involved in the streptomycin trials. From the 1920s, Hill applied statistics to medicine, attending the lectures of renowned mathematician Karl Pearson, among others. He became famous for a landmark study carried out in collaboration with Richard Doll on the correlation between smoking and lung cancer. They carried out a case-control study in 1950, which compared lung cancer patients with matched control and also began a sustained long-term prospective study into the broader issue of smoking and health, which involved studying the smoking habits and health of more than 30,000 doctors over a period of several years. His certificate for election to the Royal Society called him "the leader in the development in medicine of the precise experimental methods now used nationally and internationally in the evaluation of new therapeutic and prophylactic agents". The ethics of selecting clinical trial subjects have been controversial for much of their modern history, especially in the United States. From the 1950s up until the early 1970s, the vast majority of pharmaceutical testing in America was performed on prison populations; only after widespread abuse during testing was reported at the Holmesburg Prison did research diminish in these areas.
An initial success was achieved when medicinal chemists set out in search of the ideal SSRI with the chemical synthesis of zimelidine (figure 1) from the antihistamine drug brompheniramine, which exhibited selective inhibition of 5-HT re-uptake with minimal inhibition of norepinephrine (NE) re-uptake. Most importantly, zimelidine did not come with the adverse effect profile as the TCAs and therefore it became the template for the second generation SSRIs. Zimelidine was the first SSRI to be marketed, but several cases of Guillain–Barré syndrome were associated with the use of the drug which led to withdrawal from the market in 1983. Subsequently, several non-tricyclic SSRIs were discovered and marketed. Fluoxetine, which was FDA approved in 1987, is usually thought to be the first SSRI to be marketed. The work which eventually led to the discovery of fluoxetine began at Eli Lilly and Company in 1970 as a collaboration between Bryan Molloy and Ray Fuller. It was known at that time that the antihistamine diphenhydramine showed some antidepressant-like properties. 3-Phenoxy-3-phenylpropylamine, a compound structurally similar to diphenhydramine, was taken as a starting point. Molloy and fellow Eli Lilly chemist Klaus Schmiegel synthesized a series of dozens of its derivatives. Hoping to find a derivative inhibiting only serotonin reuptake, another Eli Lilly scientist, David T. Wong, proposed to retest the series for the in vitro reuptake of serotonin, norepinephrine and dopamine, using a technique developed by neuroscientist Solomon Snyder.

It mirrors the 44-residue form of human growth hormone-releasing hormone. A hexenoyl group on the N-terminal tyrosine distinguishes it from the unmodified hormone. The change is intended to improve resistance to enzymatic breakdown.
The N-terminal modification reduces cleavage by circulating peptidases, so the peptide persists longer than native GHRH. That persistence is the main rationale for the synthetic design. Comparative half-life values in humans are reported in regulatory review documents rather than in general reference literature.
Reductions in visceral adipose tissue have been measured in controlled studies of defined populations. Whether the effect generalizes to other groups and persists after treatment stops is less clear. Longer-term outcome data remain limited.
