=== Medication === Opioid replacement therapy (ORT), also known as opioid substitution therapy (OST), Medication for Addiction Treatment (MAT), or Medications for Opioid Use Disorder (MOUD), involves replacing an opioid, such as heroin. Commonly used drugs for ORT are methadone and buprenorphine/naloxone (Suboxone), which are taken under medical supervision. Buprenorphine/naloxone is usually preferred over methadone because of its safety profile, which is considered significantly better, primarily regarding its risk of overdose and effects on the heart (QTc prolongation). Buprenorphine/naloxone, methadone, and naltrexone are approved by the U.S. Food and Drug Administration (FDA) for medication-assisted treatment (MAT). In the U.S., the Substance Abuse and Mental Health Services Administration (SAMHSA) certifies opioid treatment programs (OTPs), where methadone can be dispensed at methadone clinics. As of 2023, the Waiver Elimination (MAT Act), also known as the "Omnibus Bill", removed the federal requirement for medical providers to obtain a waiver to prescribe buprenorphine, in an attempt to increase access to OUD treatment. == History == Habitual drunkenness was described as a medical condition well before the modern concept of addiction existed. The American physician Benjamin Rush and the British naval physician Thomas Trotter, writing independently in the late eighteenth century, are conventionally credited with consolidating the idea that compulsive drinking was a disease rather than a vice — an idea that gained wide currency only through the nineteenth century, alongside the temperance movement. Comparable claims were made for drugs somewhat later. In 1877 the Berlin physician Eduard Levinstein published Die Morphiumsucht, the first monograph to treat morphine addiction as a disorder in its own right; an English translation, Morbid Craving for Morphia, appeared the following year. Levinstein defined the condition as an uncontrollable desire to use the drug, but argued that it was not a mental illness, on the grounds that the impairment of will did not extend beyond drug use into other areas of life.

Formation of a secondary structure is the first step in the folding process that a protein takes to assume its native structure. Characteristic of secondary structure are the structures known as alpha helices and beta sheets that fold rapidly because they are stabilized by intramolecular hydrogen bonds, as was first characterized by Linus Pauling. Formation of intramolecular hydrogen bonds provides another important contribution to protein stability. α-helices are formed by hydrogen bonding of the backbone to form a spiral shape (refer to figure on the right). The β pleated sheet is a structure that forms with the backbone bending over itself to form the hydrogen bonds (as displayed in the figure to the left). The hydrogen bonds are between the amide hydrogen and carbonyl oxygen of the peptide bond. There exists anti-parallel β pleated sheets and parallel β pleated sheets where the stability of the hydrogen bonds is stronger in the anti-parallel β sheet as it hydrogen bonds with the ideal 180 degree angle compared to the slanted hydrogen bonds formed by parallel sheets.
In the Philippines, 7-Eleven was run by the Philippine Seven Corporation (PSC). Its first store, located at the corner of EDSA and Kamias Road in Quezon City, opened on February 29, 1984. On July 28, 1988, PSC transferred the Philippine area license to operate 7-Eleven stores to its affiliate, Phil-Seven Properties Corporation ("PSPC"), together with some of its store properties. In exchange thereof, PSC received 47% of PSPC stock as payment. On May 2, 1996, the stockholders of both PSC and PSPC approved the merger of the two companies to advance PSC group's expansion. On October 30, 1996, Securities and Exchange Commission approved the merger and PSPC was then absorbed by PSC as the surviving entity. In 2000, President Chain Store Corporation (PCSC) of Taiwan, also a licensee of 7-Eleven, purchased the majority shares of PSC and thus formed a strategic alliance for the convenience store industry within the area. In February 2009, 7-Eleven signed a non-exclusive contract with Chevron Philippines to open its stores in selected Caltex gas stations nationwide. In 2012, they opened their first store outside of Luzon in Cebu City, which soon expanded to the other parts of Cebu as well as its neighboring provinces. It was followed with the branch openings in Bacolod in 2013, Iloilo City in 2014, Davao City and Cagayan de Oro in 2015. The number of stores eventually spread from these major cities to smaller towns and provinces near them.

Spherocytes are slightly smaller than normal biconcave red blood cells. Mean corpuscular hemoglobin concentration (MCHC): Increased (normal range: 31–36% Hb/cell). This is secondary to less water being in the cell. MCHC can be normal when HS is combined with iron deficiency anaemia or thalassaemia. Red blood cell distribution width (RDW): Increased (normal range: 11–15%). The spherocytes create variation in the size of the red blood cells on average, thus expanding the distribution. Red blood cell count (RBC): Sometimes increased early (normal range males: 4.3–5.9 million/mm3; normal range females: 3.5–5.5 million/mm3). The loss of surface area per cells cause the body to mass-produce red blood cells. The spleen will filter spherocytes out and change this value.

In 2007 a 10-year-old male Dalmatian dog was described with acromegalic signs such as an enlarged tongue and head, widened interdental space, and thickening of skin; along with other signs of hypersomatotropism such as polyphagia, obesity, and stridor. Insulin levels were heightened and the dog had glucose intolerance; however, the dog was not diabetic as blood glucose levels were found to be normal after multiple tests. Levels of IGF-1 and growth hormone were heightened. CT imaging showed a pituitary mass which was later confirmed to be an acidophilic adenoma stained for growth hormone; however, mammary tumour-induced acromegaly is rare with few reported cases. Hypothyroidism can increase levels of growth hormone and IGF-1 in dogs. Some of the physical changes in dogs with hypothyroidism may be due to increased growth hormone concentration.
== Taste and texture == Cheaper cuts of meat with connective tissue and lean muscle fibers are suitable for stewing, as long slow cooking softens connective tissue without toughening the muscle. Slow cooking leaves gelatinized tissue in the meat, so that it may be advantageous to start with a richer liquid. The low temperature of slow-cooking makes it almost impossible to burn even food that has been cooked too long. However, some meats and most vegetables become nearly tasteless or "raggy" if over-cooked. Slow cookers do not provide sufficient heat to compensate for loss of moisture and heat due to frequent removal of the lid, e.g., to add and remove food in perpetual stews, (pot-au-feu, olla podrida). Added ingredients must be given time to cook before the food can be eaten.
=== Lactic acid hypothesis === It was once believed that lactic acid build-up was the cause of muscle fatigue. The assumption was lactic acid had a "pickling" effect on muscles, inhibiting their ability to contract. The impact of lactic acid on performance is now uncertain, it may assist or hinder muscle fatigue. Produced as a by-product of fermentation, lactic acid can increase intracellular acidity of muscles. This can lower the sensitivity of contractile apparatus to calcium ions (Ca2+) but also has the effect of increasing cytoplasmic Ca2+ concentration through an inhibition of the chemical pump that actively transports calcium out of the cell. This counters inhibiting effects of potassium ions (K+) on muscular action potentials. Lactic acid also has a negating effect on the chloride ions in the muscles, reducing their inhibition of contraction and leaving K+ as the only restricting influence on muscle contractions, though the effects of potassium are much less than if there were no lactic acid to remove the chloride ions. Ultimately, it is uncertain whether lactic acid reduces fatigue through increased intracellular calcium or increases fatigue through reduced sensitivity of contractile proteins to Ca2+.
